ABSTRACT

Objective To investigate the prevalence and genotype of thalassemia in high-risk children with thalassemia in Wuzhou, and to provide a reference for formulating strategies for the prevention and treatment of thalassemia and for reducing the incidence of thalassemia. Methods Four deletions and 3 point mutations of alpha-thalassemia and 17 point mutations of beta-thalassemia were detected and analyzed by GAP-PCR and PCR combined with reverse dot blot hybridization in children at a high risk for thalassemia in Wuzhou from 2010 to 2018. Results There were 1,421 positive cases in the first screening, and 871 cases were confirmed through genotyping, with a positive rate of 61.29%, including4 deletion types and 14 mutation types. There were 452 cases (51.89%) of α-thalassemia, 337 cases (38.69%) of β-thalassemia and 82 cases (9.41%) of combination of α-thalassemia and β-thalassemia. The common genotypes of α-thalassemia in children in Wuzhou were: --SEA/αα, --SEA/-α3.7, -α3.7/αα, --SEA/αCS αCS, αWSα/αα, --SEA/-α4.2, -α4.2/αα, and αCSα/αα. CD41-42, CD17, -28 and CD71-72 were the most common heterozygotes, while CD41-42/-28, CD41-42/IVS-2-654, and CD41-42/ CD71-72 were the most common double heterozygotes in children with β-thalassemia in Wuzhou. The most common homozygous genotypes were CD41-42 / CD41-42 and -28/-28. Conclusion In order to control thalassemia it is critical to increase investment in large-scale screening of carriers of the thalassemia mutant gene, and to prevent the birth of children with severe thalassemia.

ABSTRACT

Objective To study the role of different dose nicorandil in prevention of CIN in elderly CHD patients.Methods One hundred and twenty-one elderly CHD patients were divided into standard dose nicorandil treatment group (n=42),5 mg nicorandil treatment group (n=39) and 10 mg nicorandil treatment group (n=40).Their Scr levels,and eGFR were measured for 3 days before and after PCI.The urine KIM-1 and NGAL levels were measured before and 24 h after PCI.The incidence of CIN and adverse reactions were compared in 3 groups.Results The incidence of (IN and the eGFR ≥25% were significantly lower,the serum Scr level was significantly lower while the eGFR was significantly higher in 5 mg and 10 mg nicorandil treatment groups than in standard dose nicorandil treatment group on days 1 and 2 after PCI (P<0.05,P<0.01).The serum Scr level was significantly lower while the eGFR was significantly higher in 10 mg nicorandil treatment group than in 5 mg nicorandil treatment group on day 2 after PCI (P<0.05).The urine KIM-1 and NGAL level were significantly lower in 10 mg nicorandil treatment group than in standard dose nicorandil treatment group and 5 mg nicorandil treatment group at 24 h after PCI (2.77±0.33 μg/L vs 5.63±0.27 μg/L,4.82±0.32 μg/L,P<0.05;41.64±8.42 μg/L vs 66.51±10.72 μg/L,57.11±9.67 μg/L,P<0.05).Conclusion Different oral doses of nicorandil play a role in prevention of CIN and 10 mg nicorandil plays a greater role than standard dose nicorandil and 5 mg nicorandil in prevention of CIN in elderly CHD patients undergoing PCI.

ABSTRACT

Cavernous transformation of the portal vein (CTPV) is considered a rare condition. However, with the development of radiological technology, an increasing number of patients have been diagnosed with CTPV, thus causing wide attention in clinical practice. This article elaborates on the research advances in CTPV from etiology to treatment, discusses the etiology of portal vein thrombosis and the mechanism by which portal vein thrombosis leads to CTPV, and summarizes the clinical manifestations of CTPV and the main methods of diagnosing this disease. Besides, this article analyses various methods in treating CTPV, and believes that surgical shunts are still the most extensively used method in treating CTPV.

ABSTRACT

This study sought to reveal the difference of brain functions at resting-state between subjects with sub-health and normal controls by using the functional magnetic resonance imaging (fMRI) technology. Resting-state fMRI scans were performed on 24 subjects of sub-health and on 24 healthy controls with gender, age and education matched with the sub-health persons. Compared to the healthy controls, the sub-health group showed significantly higher regional homogeneity (ReHo) in the left post-central gyrus and the right post-central gyrus. On the other hand, the sub-health group showed significantly lower ReHo in the left superior frontal gyrus, in the right anterior cingulated cortex and ventra anterior cingulate gyrus, in the left dorsolateral frontal gyrus, and in the right middle temporal gyrus. The Significant difference in ReHo suggests that the sub-health persons have abnormalities in certain brain regions. It is proved that its specific action and meaning deserves further assessment.

ABSTRACT

Objective To observe the therapeutic effect of abdominal acupuncture combined with Chinese medicine for the treatment of functional constipation. Methods Ninety-three qi-deficiency functional constipation patients were evenly randomized into western medicine group, Chinese medicine group and combination group. The western medicine group was given oral use of Dupbalac, Chinese medicine group was given modified Buzhong Yiqi decoction, and the combination group was given abdominal acupuncture and oral use of modified Buzhong Yiqi decoction. The treatment of the three groups covered 3 months. After treatment, the therapeutic effect was evaluated accord ing to the symptom scores and the examination results of gastrointestinal kinetics. Results ( 1) The total effective rate for relieving symptoms was 100.00% in combination group, 90.32% in Chinese medicine group, and 38.71% in western medicine group, and the effect was the best in the combination group ( P<0.05). ( 2) The combination group had stronger effect on relieving dyskinesis of anal sphincter, rectal sensibility, colonic transportation than Chinese medicine group and western medicine group (P<0.05). Conclusion Abdominal acupuncture combined with Chinese medicine shows certain therapeutic effect for the treatment of qi-deficiency functional constipation, and the effect is superior to that of Chinese medicine or Dupbalac.

ABSTRACT

Objective To investigate the application value of amniotic cell culture and karyotype analysis in prenatal diagnosis . Methods 1 016 pregnant women in second trimester were subject to amniocentesis under the guidance of B-type ultrasonic inspec-tion ,and the cell culture and karyotype analysis were performed on the amniotic fluid which had been drawn out .Results Among 1 016 pregnant women ,1 011(99 .5% ) succeeded in the first operation of amniocentesis and cell culture .The detection rate of fetal chromosomal abnormal karyotype was 10 .3% (105/1 016) ,in which 8 .3% (85/1 016) of structural abnormality and 2 .0%(20/1 016) of quantity abnormality .856 cases were received follow-up .Conclusion Amniotic fluid cell culture and karyotype analy-sis is a safe and reliable method for prenatal diagnosis .

ABSTRACT

The present study used the experimental patterns of Go/No Go and no motion contingent negative variation (CNV) task into the research in order to study whether the CNV can express the implication of expectation. Through comparing the CNV under different conditions, the data collected from experiment showed that the key to evoked CNV was close to the warning signal and command signal. Whether the command signal was related to the task would impact on the amplitude of the CNV. This characteristics responses to the subjects' expectation. On this basis, CNV can be used as the electrophysiological index for the reflection of expected value in the conditions of this experiment.

OBJECTIVE@#To observe the effect of hypoxia inducible factor -1α (HIF-1α) small interfering RNA (siRNA) on the expression of HIF-1α and inducible nitric oxide synthase (iNOS) in HaCaT cells under hypoxia.@*METHODS@#HaCaT cells were divided into 4 groups: the normal control group (without any treatment), the hypoxia group (under hypoxia for 24 h), the liposome control group (HaCaT cells transfected with liposome before hypoxia treatment), the RNA interference group (HaCaT cells transfected with siRNA sequences then under hypoxia for 24 h). Real-time PCR and Western blot were utilized to determine HIF-1α and iNOS mRNA and protein expression in HaCaT cells.@*RESULTS@#There was no significant difference of the mRNA expression of HIF-1α between the hypoxia group and the normoxia group (P>0.05), but the protein expressions of HIF-1α was increased in the hypoxic group than that in the normoxia group (P<0.05). Both the mRNA and protein expression of iNOS were increased in hypoxic conditions than that in the normoxia (P<0.05). Decreases were more significant in the mRNA and protein expression of HIF-1α and iNOS in the RNA interference group than that in the liposome control group in HaCaT cells (P<0.05).@*CONCLUSION@#Hypoxia increased HIF-1α and iNOS expression in HaCaT cells and inhibition of HIF-1α expression decreased iNOS expression.

ABSTRACT

ObjectiveTo observe the effects of hypoxia inducible factor-1 alpha (HIF-1α)-targeting small interfering RNA(siRNA) on the expression of HIF-1α and vascular endothelial growth factor (VEGF) in HaCaT ceils under hypoxic conditions. MethodsHaCaT cells were cultured and divided into four groups, normal control group (without any treatment), hypoxia group (cultured under hypoxic conditions for 24 hours),liposome control group (transfected with liposome followed by hypoxic culture for 24 hours), RNA interference group (transfected with HIF-1α-targeting siRNA/liposome complexes followed by hypoxic culture for 24 hours). Fluorescence real-time quantitative PCR was utilized to determine HIF-1oα and VEGF mRNA expression in HaCaT cells, and Western blot to detect HIF-1α and VEGF protein expression. ResultsNo significant difference was observed in the mRNA expression of HIF-1α between the hypoxia group and normal control group(0.907 ± 0.032 vs. 0.878 ± 0.034, F =1.108, P > 0.05), while the expression levels of VEGF mRNA,HIF-1α and VEGF proteins were significantly higher in the hypoxia group than in the normal control group (0.935 ± 0.032 vs. 0.652 ± 0.053, 0.813 ± 0.047 vs. 0.236 ± 0.014, 0.791 ± 0.030 vs. 0.316 ± 0.013, all P <0.05). A significant decline was noted in the mRNA and protein expression levels of VEGF (0.230 ± 0.044 vs.0.978 ± 0.030, 0.213 ± 0.026 vs. 0.817 ± 0.049, both P < 0.05) and HIF-1α(0.497 ± 0.033 vs. 0.806 ±0.040, 0.249 ± 0.028 vs. 0.833 ± 0.052, both P < 0.05) in the RNA interference group than in the liposome control group. ConclusionsHypoxia may enhance the expression of HIF-1α and VEGF in HaCaT cells, and to inhibit the HIF-1α expression may suppress the expression of VEGF in HaCaT cells under hypoxia.

ABSTRACT

OBJECTIVE@#To investigate the expression of inducible nitric oxide synthase (iNOS) and hypoxia-inducible factor 1(HIF-1) α in psoriatic lesions,and to explore their relationship with angiogenesis in psoriasis.@*METHODS@#HIF-1α and iNOS protein were detected by immunohistochemistry and Western blot in 32 cases of psoriasis and 20 healthy controls,and CD34 marking vascular endothelial cells were used to measure the microvascular density (MVD).@*RESULTS@#The expressions of HIF-1α and iNOS protein were very weak in the control skin, but very strong in psoriatic lesions, which showed significant difference in the expressions of iNOS and HIF-1α between the psoriasis and the control group(P<0.05). Expression of HIF-1α (r=0.743, P<0.01) and iNOS (r=0.639,P<0.01) had positive correlation with MVD in psoriasis respectively. There was a positive correlation between iNOS and HIF-1α expression in psoriasis (r=0.717, P<0.01).@*CONCLUSION@#Both iNOS and HIF-1α have high expression in psoriasis and might play an important role in the genesis and development of psoriasis.

ABSTRACT

OBJECTIVE: To study the effect of Qi-replenishing and Yin-nourishing in inhibiting tumor metastasis. METHODS: Fifty mice were randomly divided into replenishing Qi group, replenishing Yin group, replenishing Qi and Yin group, chemotherapy group and control group. Models of Lewis lung cancer and B(16) melanoma were observed in vivo. The tumor weight inhibiting rate, metastatic lung tumor, concentrations of t-PA and TXB(2) in tumor tissue and IL-2 and TNF-alpha in spleen were observed. RESULTS: The tumor weight inhibiting rates of replenishing Qi and Yin group for Lewis lung cancer and B16 melanoma were both 42.9%. Replenishing Qi and Yin could decrease the contents of TXB(2) and 6-keto-PGF(1alpha), and increase the levels of IL-2 and TNF-alpha. CONCLUSION: Replenishing Qi and Yin can promote the healthy energy to inhibiting tumor metastasis.

ABSTRACT

Objective: To investigate the role of T cell in the antitumor immune responses induced by MIF gene-modified tumor vaccine. Methods: MIF gene was transferred into FBL3 erythroleukemia cel l by adenovirus carrier and a new type of tumor vaccine was prepared. The chang es of the number and the function of T cell in spleen and lymph node was observe d. Results: After the mice were immunized with MIF gene-m odified FBL3 vaccine, the number of lymphocyte in spleens and lymph nodes increa sed markedly and the specific CTL activities of splenocytes also increased great ly. FACS analysis showed that the CD3+, CD4+, CD8+ T cells and CD28 posi tive cells in draining lymph nodes of MIF-FBL3 group mice increased more marked ly than that of control groups. When the wild type FBL3 cells were injected into the mice immunized with MIF gene-modified FBL3 vaccine, the growth of tumors w ere obviously inhibited and the survival rate of the mice was increased. Conclusion: It is suggested that MIF gene-modified tumor vaccine can induce specific antitumor immune responses mediated by T cells and may be a candidate for gene therapy of tumor.

ABSTRACT

Objective: To study the expression and activity changes of methionine adenosyltransferase (MAT) in human peripheral T lymphocytes. Methods: The expression of MAT mRNA was detected by RT-PCR and the activity of MAT was measured. Results: After stimulated by IL-2, PHA and anti-CD3 antibody, MAT-Ⅱ gene expression increased by (8.9? 2.1), (7.7?1.9) and (8.0?1.8) times, respectively, and the expression peak was at 8, 4 and 8 h,respectively; MAT activity continuously increased in 48 h. S-adenosylmethioinie (SAM) moderately induced IL-2 and IFN secretion by human T cells. SAM(0.1 mg/ml) downregulated the expression and activity of MAT-Ⅱ and the secretion of IL-2 and IFN induced by PHA or anti-CD3 antibody in human T cells. Conclusion:MAT is involved in the activation of T lymphocytes, and high dose of SAM may also inhibit its activation through PHA and anti-CD3 antibody.

ABSTRACT

Objective:To evaluate quality of clinical trials about acupuncture and moxibustion treatment of hyperthyrodism in China. Methods:Literature of clinical trials of acupuncture and moxibustion treatment of hyperthyroidism were attained by computer and manual retrieval,and randomization,control,selection of study objects,comparability between groups,amount of samples, description and control of source of samples,criteria for diagnosis,application of blind method,criteria for assessment of therapeutic effects,statistical method,adverse effects,follow-up,and others in the literature were analyzed and evaluated by the randomized controlled test criteria and the clinical assessing method in Cochrane handbook of international Cochrane cooperation net.Results:In 63 papers,clinical randomized controlled tests of acupuncture and moxibustion treatment of hyperthyroidism only accounted for 22.2%,but with increasing tend year by year in recent years.Conclusion:Though acupuncture and moxibustion have been widely applied in prevention and treatment of hyperthyroidism,it could not provide evidences of higher reliability for clinical treatment due to less clinical randomized controlled tests and lower quality,which severely hinder testing and verifying of clinical therapeutic effects of acupuncture and moxibustion.It is proposed that multiple central and randomized controlled test should be made,so as to search for feasible acupuncture and moxibustion methods with definite therapeutic effect for hyperthyroidism,and provide basis for further systematical evaluation of acupuncture and moxibustion treatment of hyperthyroidism.

ABSTRACT

Interleukin 24(IL 24),also called the melanoma differentiation associated gene 7(MDA 7),is stably expressed in human tissues associated with the immune system such as the spleen,thymus,peripheral blood leukocytes and normal melanocytes. IL 24 binds to IL 20 and IL 22 receptor complexes(IL 22R1/IL 20R2 and IL 20R1/IL 20R2),and induces secretion of high levels of IL 6,TNF alpha,and IFN gamma and low levels of IL 1beta,IL 12,and GM CSF from human PBMC. Adenoviral IL 24(Ad IL 24) induces growth suppression and apoptosis selectively in diverse human cancers and tumor cell lines without producing any apparent harmful effect in normal cells. The effects of Ad IL 24 are associated to the ratio of pro apoptotic(BAX,BAK) to anti apoptotic(Bcl 2) proteins,and the up regulation of p38 MAPK and a family of growth arrest and DNA damage(GADD) inducible genes. These demonstrate the potential therapeutic effects of Ad IL 24 in human cancer.
